ERRC scientists will send extracted DNA from bacterial pellets, derived from in vitro culturing of the gut microbiota, to the vendor via two shipments (the first with 177 samples, the second with 133 samples, for a total of 310 samples). Shipments will occur within a 12-month period, and upon receipt, the vendor will have 45 days to complete shotgun metagenomic sequencing and transfer the data files. The vendor must use the Illumina pipeline to ensure data comparability with existing project data. The work includes DNA library generation (including fragmentation method, addition of adaptors and barcode indices, and pooling), quality control with DNA quantification or gel electrophoresis, and inclusion of both negative and positive controls during library preparation. Shallow sequencing must produce 3-5 million read pairs per sample on the Illumina platform. All raw data files must be transferred to the ERRC scientists, with quality control on reads, including demultiplexing and removal of adaptor sequences. All work must be performed in the United States. Bidders must outline all steps in their quotes and provide specific details on methods, adaptors, and controls used. Quotes must include company UEI, technical documentation with a "meets/does not meet" statement for each minimum specification, pricing with total and unit costs including shipping, an estimated delivery timeline, and validity of at least 60 days. The vendor must comply with all requirements and deliverables as specified.
